From orange-yellow to red – a new color option for anaerobic bacteria detection

In anaerobic bacteria detection, the color development of traditional indole-based substrates (such as X-Gal) depends on oxygen oxidation, resulting in weak signals under strictly anaerobic conditions and making positive results difficult to identify. GeneSeqTools Bioscience & Technology previously launched the anaerobic bacteria-β-D-galactosidase orange-yellow chromogenic substrate (GST1010), which solved the "anaerobic color development" problem. Now, the Anaerobic Bacteria-β-D-Galactosidase Substrate (Anaerobic β-D-galactosidase substrate, GST1011) extends this core technology to the red spectrum – utilizing the same intramolecular condensation color development mechanism, but with a completely new red insoluble precipitate signal, allowing clear and intuitive in-situ visualization of anaerobic bacterial colony signals in culture media.

The Anaerobic Bacteria-β-D-Galactosidase Red Substrate is a chromogenic substrate specifically designed for β-galactosidase detection under anaerobic conditions. After enzymatic cleavage, it undergoes intramolecular condensation to form a red insoluble precipitate without requiring oxygen or auxiliary reagents. It is suitable for enzyme activity detection of obligate anaerobes such as Bacteroides and Bifidobacterium, as well as facultative anaerobes such as E. coli. Available in stock with custom packaging supported, HPLC purity ≥98%.

II. Working Principle

The color development mechanism of the Anaerobic Bacteria-β-D-Galactosidase Red Substrate is consistent with that of the orange-yellow chromogenic substrate, both based on intramolecular condensation reaction – color development occurs without oxygen after enzymatic cleavage. The entire reaction process can be summarized in three stages: "cleavage – cyclization – color development":

1. Enzyme recognition and bond cleavage: β-Galactosidase specifically recognizes and hydrolyzes the β-D-galactoside bond in the substrate, releasing a colorless alditol dye precursor.

2. Intramolecular condensation: The released precursor molecule spontaneously undergoes intramolecular condensation without the need for oxygen or any auxiliary oxidant, forming a conjugated chromogenic structure.

3. Precipitation and color development: The condensation product is an insoluble red dye that precipitates in situ on the colony, helping to maintain in-situ colony color development.

Since the color development process does not rely on oxidative dimerization, stable and consistent color development results can be obtained under both aerobic and anaerobic culture conditions. Compared to traditional indole-based substrate systems that require oxygen for color development, this substrate is more suitable for applications such as anaerobic bacteria culture, screening, and identification.
